Boiler/chiller water pressure question...

Lets say I have a building that calculates out to need 40 psi in order to get water to the top of it. Would I just fill the boiler or chiller to 40 psi static pressure and call it good, or do I need to start the pumps and make sure the return pressure is above 40 psi?

Also, what calculation do people use to figure out pressure needs? If I remember correctly from school a long time ago, it's somewhere around 1/2 psi per foot. Is that correct, or am I remembering it wrong?
